"There's no doubt that it's been a difficult week with three matches in just seven days but it provides a true test of our squad depth," said Holley, who saw his side win away in Connacht on the day of the new season before defeating the Blues 32-10 at the Liberty Stadium.
"There are a lot of familiar faces in the team with a good splash of youth. This is a real indication of the good work we are doing to develop the best local young talent.
Glasgow have themselves started well, returning from Rodney Parade with a 12-6 win over the Dragons.
"We've had a solid start in the Magners League but we're expecting a real test.
"Glasgow played some exciting rugby last season and have started well this time around with a win at Rodney Parade against the Dragons last weekend.
"We are well aware that we'll have to be at our best to get a result."
New signing Jamie Nutbrown and 18-year-old Dan Biggar make their first competitive starts for the Ospreys at half-back while David Bishop plays for the first time in more than three years after suffering a fractured kneecap.
