Newport Gwent Dragons lock Luke Charteris and Neath Swansea Ospreys back row Ryan Jones are the two new faces in the squad set to win their first caps if selected during the series.
Cardiff Blues wing Craig Morgan returns to the squad and could figure in a Wales shirt for the first time since his Six Nations appearance against France in 2003, with the likes of Llanelli Scarlets Mark Jones and NECHarlequins Dafydd James currently ruled out through injury.
"It's been a difficult but exciting selection so early into the season," said Ruddock.
"We have picked 29 players but there is a degree of flexibility that could allow one or two more to come into the squad in what is a fluid and ongoing selection situation.
"There are a number of players who went on the summer tour who are unlucky to have missed out because others have returned from injury.
"But there are plenty of games between now and the start of the autumn series and there is no reason why people can't play themselves back into contention with performances at regional level.
"We are playing some of the top nations in World rugby in just over a month's time and are all very much looking forward to the challenge.
"A particular welcome must go out to Luke Charteris and Ryan Jones who have the potential to pick up their first caps during the series
.
"Both have impressed, Ryan in his regional performances this season and Luke through is career in under-age rugby and his appearances with the Dragons.
"Luke's selection was part of our planning before the summer tour, one of the reasons we left him at home was to work on his muscle mass and he certainly has the size to make an impact at international level at 6 foot 9 inches."
Ruddock is expected to name his captain for the Autumn series over the next few days after being blessed with a number of high quality candidates, but he is more than content with the current blend of his squad.
"A captain for this squad and the season ahead will be announced later this week," added Ruddock.
"In the meantime a number of high quality candidates will meet individually with the team management as part of a process that will determine the captain and to fully explore and understand the roles and responsibilities of the job.
"We are happy with the blend of experience, talent, skill and potential we have in this squad and are now looking for those who have been named today to step up to the mark and commit themselves to an exciting Autumn international series."
Ospreys' hooker Huw Bennett, Scarlets' hooker Robin McBryde and the Blues backline trio Ryan Powell and Nicky and Jamie Robinson join James and Jones on the 'ruled out through injury' list.
